Investigators examining DNA recovered from the home of Nancy Guthrie are uncertain whether the biological material will provide enough information to help identify individuals connected to her disappearance, according to people familiar with the inquiry.

Sources say the DNA sample collected inside Guthrie’s residence in Tucson, Arizona appears to be of low concentration. Law enforcement officials explained that such “low-level” DNA evidence often contains insufficient genetic material to build a clear and reliable profile suitable for comparison with national or commercial DNA databases.

While laboratory scientists can sometimes strengthen weak DNA samples using advanced processing techniques, the procedure may require additional time and does not always guarantee usable results.

The analysis is being conducted by a private laboratory in Pima County Sheriff's Department’s contract testing partner located in Florida. Officials said the testing has not yet reached a final conclusion regarding whether the material can be transformed into a full genetic profile.

As the investigation enters its fourth week, authorities have not publicly identified any suspect or person of interest. Law enforcement officials confirmed that all three of Guthrie’s adult children and their spouses were cleared after preliminary scrutiny of their potential involvement.

The family has announced a reward of up to $1 million for information leading to Guthrie’s safe return. Separately, the Federal Bureau of Investigation has offered an additional $100,000 reward in exchange for credible information.

Earlier forensic analysis of DNA collected from gloves discovered approximately two miles from Guthrie’s home did not match records stored in the FBI’s national DNA registry, known as the Combined DNA Index System.

Authorities are now relying partly on genetic genealogy methods to assist the investigation. This technique compares unknown DNA samples with data stored in public genealogy databases to identify possible relatives of an unidentified individual.

Experts say law enforcement agencies typically use consent-based platforms such as GEDmatch and FamilyTreeDNA when conducting investigative searches. Participation in such systems is voluntary, and users can choose whether their genetic data is accessible to authorities.

Representatives from FamilyTreeDNA emphasized that the company operates under strict privacy standards and does not directly conduct investigative work for police agencies. Instead, it collaborates with independent partners that manage law enforcement queries using opt-in consent databases.

Officials from GEDmatch explained that their platform allows authorities to search DNA profiles to identify criminals or unknown human remains. The company noted that law enforcement usage of the service is often disclosed publicly only after investigations produce results.

The disappearance of Guthrie has attracted national attention, partly because of her connection to television journalist Savannah Guthrie, who has publicly expressed hope for her safe return.

Investigators continue exploring multiple leads while forensic teams attempt to determine whether the DNA evidence can be refined into a usable profile. Authorities say privacy laws and technical limitations sometimes restrict how genetic data can be processed.
