Wednesday started like any other trading day for Moderna. It didn't end that way. Shares of the biotech more than doubled in a single session, tacking on roughly $30 billion in market value after promising late-stage trial results for its personalized mRNA cancer therapy — developed in partnership with pharmaceutical giant Merck — sent investors rushing to buy in.

A Bet on mRNA Beyond COVID

The therapy sits at the heart of Moderna's broader strategy: proving that its mRNA platform, best known for powering its COVID-19 vaccine, can do far more than fight infectious disease. The company has spent the past year diversifying its pipeline, recently adding a flu vaccine to its lineup while pushing forward experimental shots targeting conditions like norovirus and Lyme disease — a deliberate effort to build revenue streams that don't rise and fall with pandemic-era demand.

Analysts Saw This Coming

Wall Street watchers had flagged the cancer vaccine data as a potential turning point for the company well before Wednesday's numbers landed. "A successful trial gives investors stronger evidence that mRNA technology can have a meaningful future beyond infectious diseases," said Lale Akoner, global market strategist for eToro — a vote of confidence that, once validated by real trial data, sent the stock rocketing.

Peter Andersen, founder of Andersen Capital Management in Boston, said much of the excitement stems from investors trying to price in the treatment's future commercial potential. "Investors are probably trying to estimate the gain in revenue and earnings from such a medication if it does exist," Andersen said, adding that a genuinely effective treatment for a common cancer could prove "extremely profitable for the company."

The Trial Results Behind the Surge

The vaccine, known as intismeran, showed it could reduce the risk of skin cancer recurrence and spread when paired with Merck's blockbuster immunotherapy drug Keytruda, according to results from a late-stage trial — the kind of data that can transform a speculative pipeline bet into a near-certain commercial product almost overnight.

A Rising Tide Across Biotech

Moderna's surge didn't happen in isolation — it lifted the entire sector with it. The Nasdaq biotech index climbed 4.4% to a fresh record high, buoyed largely by Moderna's dramatic gains. U.S.-listed shares of BioNTech jumped 20%, while Novavax added 6%, as investors extended their optimism about mRNA technology across the board, treating Moderna's win as validation for the whole approach.

The Numbers Tell the Story

Moderna's shares hit a two-year high of $140.19, putting the company on pace to add roughly $30 billion to its market capitalization in a single trading session. Trading volume exploded to more than 15 times the stock's 50-day average, according to data from LSEG — a surge that dwarfed typical daily activity by an order of magnitude. Merck rode the wave too, climbing 10% to an all-time high of its own.

Analysts Raise Their Sales Forecasts

The excitement wasn't just sentiment — it came with hard numbers attached. Leerink Partners analysts revised their sales projections for the therapy upward, now estimating about $1.4 billion in potential sales by 2032, up from a previous ceiling of roughly $1.2 billion.

A Short Squeeze Adds Extra Fuel

Beyond the trial data itself, market mechanics may have amplified the rally even further. "With each shorted share now down almost $100, that is exactly the kind of pressure that can trigger a squeeze," said Peter Hillerberg, co-founder of ORTEX, describing a dynamic where investors betting against the stock suddenly found themselves scrambling to limit their losses.

"There is clearly a real catalyst behind a move this size, but with a short base this large, shorts scrambling to cover are adding fuel and can keep driving the price higher," Hillerberg added — a feedback loop where panic-buying from short sellers can push a stock even higher than the underlying news alone might justify.

Just How Crowded Was the Short Trade?

The numbers help explain why the squeeze hit so hard. Short interest in Moderna's shares stood at 13.5% of the stock's free float as of Wednesday, according to ORTEX — a level that made Moderna one of the most heavily shorted large-cap stocks on the market, and primed it for exactly the kind of explosive move it just delivered.
